• If you have 19mm male headers (instead of typical 11mm headers) and a male to female 6-pin cable, you do not need to desolder the SPI connector on the Arduino Due.

• Plug the female 6-pin connector to the SPI connector near the center of the Arduino Due.

• 

• The taller headers will raise the Arduino high enough so there is clearance for the SPI cable.
